AB Background: Experimental transmission of BSE to cynomolgus monkeys is an animal model for variant Creutzfeldt-Jakob disease (vCJD). It is most likely that vCJD was caused by the consumption of BSE-contaminated food.
Aims: We examined the distribution of PrPres in the brains of primates infected with BSE to determine the time course and to examine the role of the inoculation route on the distribution of PrPres in the brain.
Methods: Macaques were infected either intracerebrally (5 mg) or orally (5g) with BSE brain homogenate. A number of orally inoculated macaques were sacrificed at regular intervals (1, 3 and 5 years post infection), and others were sacrificed showing 14-3-3-positive cerebrospinal fluid samples only, at the onset of mild clinical signs or during the terminal stage of the disease. PrPres deposits were detected by Western immunoblot and by Petblot technique.
Results: PrPres deposits in the brain were not found during the incubation period. There were significant differences in the distribution of PrPres between intracerebrally and orally infected macaques. In orally dosed macaques, the BSE-inducing agent may have entered the brain via the medulla oblongata although, other ways cannot be completely ruled out.
Conclusions: The distribution of PrPres deposits in the brain is affected by the inoculation route.PrPres detection in the brain is a late event in simian vCJD. However, pathological changes could be detected very earlier during the incubation period.
