AB During active surveillance of the UK sheep population for TSEs, about half the suspect cases identified by rapid testing for abnormal PrP in the brain stem were found to be distinct from the classical form of scrapie. Theses cases appear to have PrP with a less biochemically stable core structure and under stringent proteolysis conditions PrP was no longer detectable by ELISA or Western Blot (Everest et al., 2006). Mild conditions of proteolysis, however, identified a molecular banding profile on Western Blot that was distinct from the classical scrapie samples and which bore similarities with Nor-98 and similar cases now found in Europe. In addition, this newly identified variant of scrapie appears to favour sheep carrying alleles for the AHQ and ARR (and AF141RQ) genotypes. We have extended these studies to determine the biochemistry of the prion abnormality in atypical scrapie samples from a range of sheep genotypes by comparing the kinetics of proteolysis with those with prion protein from normal sheep brain and confirmed cases of classical scrapie. ELISA and Western Blot have been used to identify the molecular characteristics of the products of digestion. These studies will facilitate our understanding of the prion characteristics in the UK and underpin strategies for the control and eradication of TSEs in small ruminants.
