AB The use of PrPsc as a pre-clinical, or general marker for surveillance is limited, due to the fact that it is present in extremely small amounts in accessible tissues, or in body fluids such as cerebrospinal fluid (CSF), blood and urine. Specific detection of these small amounts of the PrPsc conformer are further exacerbated by the presence of a large excess of endogenous PrP. Thus the identification of alternative biomarkers applicable to the development of diagnostic tests or intervention therapies are needed. In this study weekly collections of body fluids were performed on ME-7 infected C57/BL6 mice and age matched controls. The extracted proteins were analyzed by 2-D fluorescence difference gel electrophoresis (2D-DIGE). Differential in gel analysis using DeCyder software (GE Healthcare) was used to identify differentially expressed proteins and gel spots of interest were isolated and identified by tandem mass spectrometry. Currently, 2D-DIGE and Western analysis are being used to characterize the protein profiles of ME-7 and age matched controls throughout disease progression.
