AB We had developed a proof method for contamination and remaining contamination on surgical instruments known as "radionuclide method". After a mock contamination of the instruments with radioactively labelled blood, the instruments are to be viewed on a gamma camera before and after the cleaning. That provides a simple and a non destructive way to localize and measure the remaining contamination both pin-pointedly and quantitatively, which is especially useful for study of reprocessing those instruments with channels and cavities such as endoscopic instruments.
Pathological prion protein, the agent that causes transmissible spongiform encephalopathies (TSE), is found to be able to bind to surgical steel surface and hard to be cleaned away; therefore, which may pose a potential threat of iatrogenic transmission. In searching of new chemical and physical decontamination procedures, question arised has been whether the prion proteins are washed away, denatured or destroyed from surface of an instrument. To answer the question we adapted "radionuclide method" in such a way that purified pathological prion proteins are radioactively labelled and the labelled prion proteins will be used as soil to contaminate test instrument. For the first step the test instrument shall be surgical steel wires. After reprocessing and remaining gamma count measuring, the wires will be used later in a Bioassay to see whether there is any correlation between gamma counts and surviving time of the test animals. If a correlation could be established, then the answer could be found about the nature of a reprocessing procedure against pathological prion proteins on the surface of the test instrument.
