AB There is some evidence to indicate that the mechanism of neuronal cell death in prion diseases is apoptosis. In vitro studies have demonstrated a relationship between the PrPsc accumulation and the induction of apoptosis via mitochondria. This pathway is regulated by expression of the proto-oncogene Bcl-2 family including Bax and Bcl-2. Nevertheless, in vivo investigations show contradictory results. In the present study, we have analysed medulla oblongata from natural scrapie infected sheep as a model of in vivo prion diseases. Our research is focused in the analysis of Bax and Bcl-2 expression and caspase-3 activation and their relationships with PrPsc accumulation, spongiosis and vacuolization. We detected the increase of Bax mRNA and protein levels and the correlation between Bax staining and scrapie histopathological features and therefore with PrPsc accumulation. Moreover, we observed an increase in the activated form of caspase-3 in neurons from scrapie animals which was negatively correlated with Bax expression and independent from prion deposition, vacuolization or spongiosis. This work analyses the involvement of the mitochondrial pathway in the neuronal death observed in a model of natural prion diseases.
