AB The susceptibility of neurons versus glial cells for exposure to scrapie was examined in cultures of dorsal root ganglia (DRG) from embryonic mice. Several glial cells (Schwann or satellite cells), but no neurons, contained granules immunopositive for scrapie at 3 days after exposure to homogenates of scrapie-infected GT1-1 cells. At 6 days after exposure and later, immunolabeled granules could no longer be detected in glial cells, while after 11 days a few neurons showed immunopositive granules in their cytoplasm. After incubation with a lysosomal enzyme inhibitor, leupeptin, the clearance of scrapie immunopositive granules from the glial cells was inhibited. However, incubation with leupeptin had no effect on accumulation of scrapie immunopositive granules in neurons. Co-cultures of DRG neurons with scrapie-infected GT1-1 cells or dendritic cells exposed to scrapie were also investigated. Our observations indicate that non-neuronal cells such as glial cells or dendritic cells, as opposed to neurons, can readily take up scrapie, which thereafter may be degraded by lysosomal enzymes, and that the relative resistance of neurons to accumulate scrapie is not the result of a higher leupeptin-dependent lysosomal enzymatic activity in these cells as compared to glial cells.

IN Die Autoren nahmen Neuronen und Gliazellen aus dorsalen Spinalganglien von Mäusen in Zellkultur und gaben Homogenate Scrapie-infizierter Zellen hinzu. Viel schneller als die Neuronen nahmen Gliazellen das PrPsc auf, bauten es aber zunehmend wieder ab, bis es in ihnen nicht mehr nachweisbar war. Im Gegensatz dazu nahm die Menge des PrPsc in die Neuronen zu und verschwand nicht mehr.
