AB Aquaporins (AQPs) mediate water flux through the plasma memebrane and play a critical role in maintenance of homeostasis of cells. AQPs water channels have an hourglass form, the narrowest segment of which permits the passage of water but not of protons and other cations under the control of size restriction and electrostatic repulsion. AQPs are different expressed in several cell types. AQP1, AQP4, AQP5 and AQP9 are AQPs present in brain. Previous studies have shown that AQPs play important roles in the regulation of brain edema following traumatic injury, cerebral ischemia, hyponatremia, and in brain tumours. It is conceivable that AQPs are crucial in water transport not only in physiological conditions but also in pathological scenarios with homeostasis alterations. Vacuolization or spongiform change is, probably, the most characteristic feature in prion diseases. Spongiform change consists in the formation of rounded vacuoles within the neuropil. When many of those vacuoles become confluent it results in large vacuoles distorting the cortical cytoarchitecture. It can be hypothesized that vacuolization is associated with changes in homeostasis in cell or with changes in water transport across cell membrane. Based on this hypothesis, we studied AQP1 and AQP4 expression in frontal cortex of 10 patients with CJD, and, on the other hand, in two transgenic mice infected with scrapie and BSE, respectively. The results show increased levels of AQP1 and AQP4 in every case.
