AB Efforts to develop assays for the blood-based ante-mortem diagnosis of TSEs have been hampered by the extremely low amounts of the pathological isoform of the prion protein (PrPsc) in body fluids. In an attempt to devise an assay that could achieve the necessary sensitivity, we have developed a method for detecting prion protein fibrils that takes advantage of kinetic differences between seeded and unseeded polymerization of prion protein monomers. In the presence of prion fibril seeds the association of labelled recombinant PrP monomers in plasma proceeds much more efficiently than in the absence of seeds. The labelled prion protein fibrils are then detected by flow cytometry. Using this method we were able to detect synthetic prion protein fibrils added to plasma down to attomolar concentrations. Preliminary results with serum from BSE-positive cattle indicate that our method may be sufficiently sensitive for the blood-based diagnosis of BSE.
