AB In scrapie-infected animals, the presence of low levels of infectivity in blood has been demonstrated in the absence of PrPsc detectable by current methods. The pattern of PrPc expression on blood cells may help to determine which cells become infected and/or facilitate dissemination of infectivity. PrPc expression on different blood fractions from uninfected sheep was investigated by flow cytometry and Western blotting. PrPc was detected on PBMC, but not granulocytes, red blood cells or platelets by flow cytometry. However, using Western blotting PrPc was also detected in the platelet fraction, suggesting that it may be found intracellularly in these cells. Four PrP-specific monoclonal antibodies with different epitopes were used to analyse the expression of PrPc on PBMC from uninfected sheep with different genotypes using flow cytometry. Our preliminary results suggest that VRQ/VRQ animals show higher levels of PrP expression with each antibody compared to ARQ/ARQ and ARR/ARR animals, and we are repeating the experiment on larger numbers of sheep. The distribution of PrPc was also analysed on different subsets of PBMC isolated from blood of clinically-affected scrapie sheep and healthy sheep with genotypes VRQ/VRQ and VRQ/ARQ, by two-colour flow cytometry. PrPc expression was detected on all the cell subsets examined, with the highest levels of expression found consistently on CD21+ and CD62L+ cells. No significant differences were found between scrapie-affected and unaffected sheep.
