AB BSE in cattle has been linked to the human TSE, variant Creutzfeldt-Jakob disease (vCJD). In the UK, it is possible that some sheep had become infected with the BSE agent through consumption of meat and bone meal (MBM) prior to the ban of July 1988 on the inclusion of ruminant derived MBM in concentrate feed for ruminants. Sheep experimentally exposed to oral challenge with the BSE agent have been shown to be susceptible to BSE infection and this reinforces the probability that sheep could have become infected via exposure to contaminated feed.
VLA has developed two immuno-based test systems, Western immunoblotting (WB) and immunohistochemistry (IHC), and these have been shown to discriminate between natural scrapie in sheep and sheep which have been experimentally infected with bovine BSE. If BSE has entered the UK sheep flock during the course of the cattle BSE epidemic it is likely that it exists in sheep several generations on from the original infection. For the study presented here, BSE brain tissue from primary infected sheep (oral challenge) was orally fed to unaffected sheep or inoculated intra-cerebrally into unaffected sheep of the same breed and genotype. The results show that the differences originally observed in the primary BSE in sheep cases, for the PrPsc molecular weight signatures (WB) and PrPd distribution profiles (IHC), are maintained on secondary passage.
