A panel of MAbs was raised both to recombinant bovine PrP (aa 25-231) and to bovine synthetic peptide 153-172. Their reactivity towards PrPc from bovine, sheep, goat, human, mouse, hamster, cat, dog, rabbit, swine and PrPsc from bovine, sheep, goat and hamster (strain 263K) was assessed by WB and IHC performed without proteinase K (PK) treatment. Here we present results about 10 MAbs according to their partial mapping on PrP.
Octapeptide sequence: we selected 3 MAbs that react with all the PrPc tested but the PrPc from cat. They recognize scrapie PrPsc from sheep, goat and hamster but not BSE PrPsc. This is because their epitopes are located at the border of the PrP PK-resistant polypeptide that is slightly different between PrPsc of sheep and bovine. In IHC MAbs recognize PrPsc from infected cattle, sheep, goat and hamster.
Sequence 104-156: 2 MAbs were selected that recognize PrPc from cattle, sheep, goat and human. They detect BSE and scrapie PrPsc but not PrPsc from hamster scrapie both in WB and IHC.
Sequence 153-172: among those produced against the synthetic peptide we selected 2 MAbs that recognize only bovine and human PrPc, as well as their PrPsc isoforms both in WB and IHC. This is probably due to the presence of histidine at position 166 that is substituted with tyrosine in PrPc from other species. To note that the epitopes of these MAbs lie between the epitope of 6H4 Prionics and the recently discovered epitope YYR (Nat. Med. 2003). A third MAb recognizes PrPc from sheep, goat, and cat only as well as PrPsc from sheep and goat in WB but when tested in IHC it detects bovine PrPsc too. This suggests that the exposition of the epitope on bovine PrPc and PrPsc is affected by amino acid differences outside the epitope and by the method used to test the reactivity.
Sequence 199-218: 1 MAb reacts to all PrPc but those of cat and swine and it also recognizes BSE and Scrapie PrPsc in WB; in IHC it can only detect BSE and hamster scrapie PrPsc.
