AB We have shown that it is possible to transmit bovine spongiform encephalopathy (BSE) to a sheep by transfusion with whole blood taken from another sheep during the symptom-free phase of an experimental BSE infection. BSE and variant Creutzfeldt-Jakob disease (vCJD) In human beings are caused by the same infectious agent, and the sheep-BSE experimental model has a similar pathogenesis to that of human vCJD. Although UK blood transfusions are leucodepleted - a possible protective measure against any risk from blood transmission - this report suggests that blood donated by symptom-free vCJD-infected human beings may represent a risk of spread of vCJD infection among the human population of the UK.
IN Britische Cheviot-Schafe wurden mit je 5 g BSE-infizierten Rinderhirns gefüttert. Blut wurde derart oral inokulierten Schafen entnommen und aus Neuseeland importierten und isoliert gehaltenen Scrapie-freien Schafen mit dem für besonders kurze Inkubationszeiten bekannten Genotyp Ala/Ala am Codon 136 und Gln/Gln am Codon 171 intravenös verabreicht. Eines dieser Empfängertiere von insgesamt 19 durchgeführten Bluttransfusionen erkrankte 610 Tage nach einer Transfusion von 400 ml Blut eines 318 Tage zuvor oral mit BSE inokulierten Schafes an BSE-induziertem Scrapie. Das Spenderschaf erkrankte nach 629 Tagen an BSE-induziertem Scrapie. Obwohl das Empfängerschaf nicht direkt mit BSE infiziert wurde, entsprach das Glykosylierungsmuster seines proteaseresistenten Prionproteins dem experimenteller BSE-Infektionen bei Schafen und nicht den von anderen Scrapie-Varianten bekannten Mustern.
